Tissue-specific regulation of apoptosis-inducing factor (AIF) in response to oxidative stress.
Apoptosis-inducing factor (AIF) is a unique mitochondria- and caspase-independent apoptosis effector implicated in apoptosis and cell death induced by oxidative stress. The development of AIF knockout mice has provided conclusive evidence for its crucial physiological role in cell survival. In this study, the regulation of AIF in response to oxidative stress in lung, heart, spleen, liver, brain, and kidney was investigated. On the basis of the activity of cleaved-poly (ADP-ribose) polymerase (PARP), a caspase-mediated apoptosis, we revealed that a single dose of H(2)O(2) induced the appearance of active caspase-3 in the brain and kidney but not in the lung, heart, spleen, or liver of wild-type mice. In contrast, AIF knockout mice were partially protected against the increase of cleaved-PARP and the appearance of caspase-3 in the brain and kidney. In addition, the level of AIF protein was increased upon exposure to H(2)O(2) or o-nitrosodialkylamine in the lungs, heart, spleen, liver, and brain of wild-type mice, but not in the kidney.
